This bioenergetic analysis allows testing multiple conditions per assay well and direct, simultaneous comparisons across wells, enabling faster throughput with smaller samples compared with traditional respirometry methods. There is also a Seahorse XFp, which is ideal for experiments involving only a few samples or with limiting amounts of material. Like the XFe24 and XFe96, the XFp can monitor oxygen consumption rate and extracellular acidification rate. It is ideal for pairwise comparisons, bioenergetic phenotyping and analysis of patient-derived samples (e.g., biopsies). The Seahorse work area (250 square feet total) includes a biosafety cabinet and tissue culture incubator for cell culture plates to be used with the Seahorse instruments. Core services are accessed on a fee-for-service basis and initial consultations are available at no charge. We are pleased to begin offering services for mouse cardiac phenotyping, which includes select surgical procedures and echocardiogram assessment of outcomes. The MitoPlex is a mass spectrometry-based quantitation of approximately 30 proteins in glycolysis and oxidative phosphorylation (see diagram below) that can complement metabolomics and Seahorse respirometry to provide a detailed portrait of bioenergetic pathways in cells and tissues. Message from the Core Director The Mitochondria Metabolism and Cardiac Phenotyping Core provides comprehensive services for analysis of metabolism and mitochondrial function in the research setting. New services include central carbon chain metabolomics and a mass spec-based quantitation of approximately 30 proteins involved in glycolysis and oxidative phosphorylation, the MitoPlex (available for mouse and human in June 2018). The core team hopes to create a paradigm shift in which medicine is approached with an awareness of organelle dysfunction underlying organ dysfunction. Additionally, we are now offering mouse cardiac phenotyping services including echocardiography and surgical models including myocardial infarction and pressure overload. Roberta A. Ordering Supplies Seahorse Respirometry Seahorse Seahorse seeding plates, calibrant and flux pack cartridges are included in the per-plate charge. We highly recommended that users utilize flux packs provided by the core to ensure excess cartridges do not expire. XF base media and chemical reagents for basic mitochondrial stress tests (oligomycin, FCCP, rotenone and antimycin) are available from the core; however, we recommend that frequent users maintain their own chemical stocks to ensure experimental consistency.
